Overview

ATN161 is a five-amino acid peptide (acetyl-Pro-His-Ser-Cys-Asn-NH2) derived from the synergy region of fibronectin. It functions as a non-RGD-based antagonist of the integrin alpha-5 beta-1 (α5β1), a protein involved in cell adhesion, migration, and angiogenesis. By inhibiting α5β1, ATN161 disrupts the signaling pathways necessary for tumor-induced blood vessel formation and the survival of endothelial cells. Developed by Attenuon, it has reached Phase II clinical trials for advanced renal cell carcinoma and malignant glioma. Recent research also suggests its potential in treating acute myeloid leukemia (AML) by inhibiting the malignant proliferation and infiltration of leukemic cells, particularly when used in combination with chemotherapy.
