Overview

ATNM-400 is a novel, first-in-class antibody radioconjugate developed by Actinium Pharmaceuticals, Inc. It utilizes Actinium-225 (Ac-225), an alpha-emitter radioisotope, conjugated to an antibody via p-SCN-Bn-DOTA. The drug targets an unspecified protein that is associated with poor prognosis, is overexpressed in various cancers including non-small cell lung cancer (NSCLC), prostate cancer, and breast cancer, and is linked to increased metastasis and resistance to treatments like EGFR tyrosine kinase inhibitors (TKIs). ATNM-400 is designed to deliver potent alpha-particle radiation directly to cancer cells, inducing irreparable double-strand DNA breaks, thereby overcoming resistance to existing therapies such as Osimertinib in NSCLC and androgen receptor pathway inhibitors (ARPIs) and PSMA-targeted radiotherapies in prostate cancer. It has demonstrated strong anti-tumor efficacy and favorable tolerability in preclinical models across multiple cancer subtypes.
