Overview

KI-3106 (marketed as Atonezet) is a fixed-dose combination (FDC) therapy developed by Kuhnil Pharmaceutical for the treatment of hyperlipidemia, specifically primary hypercholesterolemia. It combines two lipid-lowering agents with complementary mechanisms: atorvastatin and ezetimibe. Atorvastatin is an HMG-CoA reductase inhibitor that reduces endogenous cholesterol synthesis in the liver, leading to an upregulation of LDL receptors and increased clearance of LDL-C from the plasma. Ezetimibe is a selective inhibitor of the Niemann-Pick C1-like 1 (NPC1L1) protein, which is the primary transporter responsible for the intestinal absorption of dietary and biliary cholesterol. By targeting both the production and absorption pathways of cholesterol, KI-3106 provides a more significant reduction in low-density lipoprotein cholesterol (LDL-C) levels than statin monotherapy. The drug was approved and launched in South Korea as an incrementally modified drug (IMD).
