ATOR-1144 is a first-in-class bispecific IgG1 antibody developed by Alligator Bioscience for the treatment of solid tumors and hematological malignancies. It simultaneously targets the checkpoint inhibitor CTLA-4 (cytotoxic T-lymphocyte antigen 4) and the co-stimulatory receptor GITR (Glucocorticoid-Induced TNFR family Related). The drug is designed to localize its activity within the tumor microenvironment, working through multiple mechanisms including the activation of effector T cells and natural killer (NK) cells, as well as the depletion of regulatory T cells (Tregs). By combining CTLA-4 blockade with GITR agonism, ATOR-1144 aims to enhance anti-tumor immune responses while potentially reducing systemic toxicity associated with systemic checkpoint inhibition.
