ATOR-4066 is a preclinical, first-in-class bispecific antibody developed by Alligator Bioscience using the proprietary Neo-X-Prime™ technology. It targets both CD40 (a co-stimulatory protein found on antigen-presenting cells) and CEACAM5 (carcinoembryonic antigen-related cell adhesion molecule 5), a tumor-associated antigen highly expressed in certain cancers such as colorectal cancer but minimally present in normal tissues. By binding to these two targets, ATOR-4066 aims to induce potent anti-tumor immune responses by selectively activating dendritic cells and T cells within the tumor microenvironment, leading to remodeling of the immune landscape and enhanced tumor cell killing. Preclinical studies have demonstrated that its activity is dependent on CEA expression in tumors and that it can eliminate large tumors with heterogeneous CEA expression, potentially limiting mechanisms of tumor escape. The drug has shown both myeloid cell-dependent and T-cell dependent anti-tumor activity in preclinical models, supporting its potential as a monotherapy or in combination with other anticancer agents such as immune checkpoint inhibitors[1][2][3][5][6].
