Overview

**Atorvastatin + colchicine** is a combination of two small-molecule drugs: atorvastatin, a statin used primarily for lipid lowering and cardiovascular risk reduction, and colchicine, an anti-inflammatory agent mainly used to treat gout and pericarditis. Atorvastatin works via competitive inhibition of HMG-CoA reductase, the rate-limiting enzyme in cholesterol biosynthesis, resulting in decreased cholesterol production and reduced risk of atherosclerotic events. Colchicine acts primarily by binding to tubulin, inhibiting microtubule polymerization and thus modulating leukocyte activity and inflammatory responses. This combination is of particular clinical interest in patients with both cardiovascular disease and indications for colchicine (e.g., gout, pericarditis) or following recent evidence for colchicine in atherosclerotic risk reduction. However, both drugs are metabolized by CYP3A4 and are substrates for P-glycoprotein, leading to significant drug-drug interaction risk, notably increased likelihood of myopathy and rhabdomyolysis due to elevated plasma levels when administered together[1][2][3].
