Atorvastatin + temozolomide is an investigational combination therapy being studied primarily for the treatment of glioblastoma, a highly aggressive primary brain tumor in adults. Atorvastatin is a statin class small molecule that inhibits HMG-CoA reductase, reducing cholesterol synthesis and interfering with protein prenylation, particularly affecting Ras signaling pathways implicated in cancer cell survival and proliferation. Temozolomide is an oral alkylating agent that induces DNA damage leading to apoptosis in rapidly dividing tumor cells. Preclinical studies have shown that atorvastatin can enhance the antitumor efficacy of temozolomide by inhibiting Ras prenylation and downstream signaling (including Erk, rS6, eIF4E), resulting in increased cancer cell death[3]. Clinical trials are evaluating this combination alongside standard radiotherapy to determine its impact on progression-free survival and overall survival in patients with newly diagnosed glioblastoma[1][2][5].
