Atorvastatin-loaded eugenol-enriched PEGylated cubosomal in-situ gel is an experimental topical formulation developed by researchers at Misr University for Science and Technology for the treatment of moderate periodontitis. This novel delivery system repurposes atorvastatin calcium, a well-established HMG-CoA reductase inhibitor, for its pleiotropic anti-inflammatory and bone-sparing properties within the oral cavity. The formulation utilizes PEGylated cubosomes—liquid crystalline nanoparticles—enriched with eugenol to enhance stability and therapeutic efficacy. These nanoparticles are incorporated into a syringeable in-situ gel designed for intra-pocket delivery, which transitions from a liquid to a semi-solid state upon contact with the periodontal environment to ensure sustained drug release. A Phase 1 clinical trial has demonstrated that this formulation, when used as an adjunct to scaling and debridement, significantly reduces probing depth, bleeding, and gingival inflammatory markers.
