Overview

ATP128 is a self-adjuvanted chimeric recombinant protein therapeutic cancer vaccine developed using the proprietary KISIMA immunization platform. It is designed to treat colorectal cancer by stimulating anti-tumor immunity through both humoral and cellular immune responses. The construct includes three main components: a cell penetrating peptide (CPP) for efficient antigen delivery, a toll-like receptor (TLR) agonist-derived peptide for self-adjuvanticity, and a multiple antigenic domain containing three tumor-associated antigens highly expressed in colorectal cancer tissues. Mechanistically, ATP128 activates NF-κB and IRF3 pathways via TLR2 and TLR4 on monocytes and dendritic cells, leading to pro-inflammatory cytokine release, type I interferon production, upregulation of costimulatory molecules, and increased MHC class I expression—thereby promoting robust T cell-mediated adaptive immune responses[1][3][6]. The vaccine has been evaluated as monotherapy or in combination with checkpoint inhibitors such as BI 754091 (ezabenlimab), particularly in patients with advanced or metastatic microsatellite stable (MSS) colorectal cancer[4][5].
