ATP152 is a therapeutic cancer vaccine candidate developed by AMAL Therapeutics (a subsidiary of Boehringer Ingelheim) using the proprietary KISIMA platform. It is a recombinant fusion protein designed for the treatment of pancreatic ductal adenocarcinoma (PDAC). The KISIMA platform construct typically consists of three functional components: a cell-penetrating peptide (CPP) for efficient antigen delivery, a Toll-like receptor (TLR) peptide agonist that acts as a self-adjuvant, and a multi-antigenic cargo containing tumor-associated antigens specific to the target cancer. In clinical development, ATP152 was evaluated as part of a heterologous prime-boost regimen alongside other KISIMA-based proteins (ATP150 and ATP162) and the oncolytic virus VSV-GP154, often in combination with the PD-1 inhibitor ezabenlimab. The program aimed to stimulate robust T-cell responses in "cold" pancreatic tumors, though the Phase 1b clinical trial (NCT04913272) was terminated.
