Overview

ATPIF1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of ATPase inhibitory factor 1 (ATPIF1), a 12 kDa mitochondrial protein that inhibits the F1 domain of the F1Fo-ATP synthase. In various cancers, including glioblastoma (GBM), ATPIF1 is overexpressed and contributes to oncogenesis by reversing the function of the mitochondrial ATP synthase complex and promoting metabolic reprogramming. Silencing ATPIF1 with siRNA in GBM cell lines (such as U-87 and U251) has been shown to decrease the expression of ATP synthase subunits, increase reactive oxygen species (ROS) levels, and trigger Caspase 3/7-mediated apoptosis. Furthermore, ATPIF1 knockdown induces cell cycle arrest at the G0/G1 stage and shifts the metabolic profile toward increased proton leak and altered extracellular acidification. This research, primarily conducted by investigators at the University of Illinois and Pennsylvania State University, highlights ATPIF1 as a potential therapeutic target for glioblastoma.
