ATR siRNA refers to small interfering RNA molecules designed to silence the expression of the Ataxia telangiectasia and rad3-related (ATR) kinase. ATR is a critical DNA damage response (DDR) kinase that coordinates the cellular response to DNA replication stress and damage. In oncological research, particularly in glioblastoma (GBM), ATR siRNA is utilized to investigate the role of ATR in tumor cell invasion, motility, and macropinocytic vesicle trafficking. By knocking down ATR expression, researchers have demonstrated defects in cell velocity and integrin internalisation, suggesting that ATR facilitates malignant cell invasion beyond its traditional role in DNA repair. While small molecule ATR inhibitors are in clinical development, ATR siRNA is primarily employed as a research tool to validate the therapeutic potential of targeting the ATR pathway.
