ATR12-351 is a genetically engineered strain of *Staphylococcus epidermidis* developed as a topical therapy for Netherton syndrome and related congenital ichthyoses. The bacterium is modified to express and deliver an active subunit of the human lympho-epithelial Kazal-type-related inhibitor (LEKTI) protein directly into the skin. LEKTI deficiency, caused by mutations in the *SPINK5* gene, underlies Netherton syndrome—a rare and severe genetic skin disorder characterized by chronic inflammation and barrier dysfunction. By restoring functional LEKTI at the site of disease through bacterial delivery, ATR12-351 aims to inhibit excessive activity of kallikrein-related peptidase 5 (KLK5), reduce inflammatory cytokines such as IL-36γ, and address the root cause of disease pathology. The drug is being developed by Azitra in collaboration with The Jackson Laboratory and has entered Phase 1b clinical trials for adult patients with Netherton syndrome[3][4][5][6][7][8].
