ATRC-101 is an engineered, fully human monoclonal antibody developed by Atreca for the treatment of solid tumors. It targets a novel, tumor-specific extracellular ribonucleoprotein (RNP) complex, with PABP-1 as its most prominent component. Upon binding to its RNP antigen on tumor cells, ATRC-101 activates the innate immune system and alters the local tumor microenvironment, promoting T-cell-mediated killing of tumor cells. Preclinical studies show that ATRC-101 increases conventional dendritic cell type 1 (cDC1), CD8+ T cells, and natural killer (NK) cells in blood and tumors while decreasing immunosuppressive cell populations such as cDC2 and monocytic myeloid-derived suppressor cells. This leads to both innate and adaptive antitumor immune responses. Clinical trials have demonstrated anti-tumor activity in patients with advanced solid tumors including non-small cell lung cancer (NSCLC) and melanoma.
