Overview

ATS-907 is a small molecule Rho kinase (ROCK) inhibitor developed for topical ocular administration, primarily targeting the reduction of intraocular pressure (IOP) in patients with primary open angle glaucoma and ocular hypertension. The drug acts as a prodrug, exhibiting excellent ocular surface penetration and rapid conversion to an active form in the anterior chamber after topical dosing, which may enhance its therapeutic index. Its mechanism involves inhibition of ROCK enzymes, leading to relaxation of trabecular meshwork cells and increased aqueous humor outflow, thereby lowering IOP. Originally discovered by Asahi Kasei Pharma and later licensed to Altheos for development outside Japan and Korea, ATS-907 advanced through Phase 2 clinical trials but was ultimately discontinued before approval[1][3][5][6][7].
