An **attenuated reassortant lymphocytic choriomeningitis virus** is an engineered, live, replication-competent or -deficient viral vector derived from LCMV exhibiting reduced virulence due to genetic modifications such as segment reassortment or codon deoptimization. These modifications result in reduction of viral protein expression or altered segment structure, substantially lowering pathogenicity but preserving immunogenicity. Such viruses have been developed as vaccine candidates, able to confer protective immunity against wild-type LCMV challenge in animal models, and engage robust cellular and antibody responses. Genome rearrangement (e.g. swapping genes or recoding nucleoprotein using rare codons) is used to achieve attenuation. Preclinical and early-phase clinical work demonstrates efficacy and safety for infectious disease prevention and immunotherapy platforms[1][5][7].
