attIL12-T cell therapy is an innovative immunotherapeutic approach that uses tumor infiltrating lymphocytes (TIL) genetically engineered with a cell membrane-anchored and tumor-targeted interleukin-12 (IL-12). This therapy is designed to overcome the limitations of conventional IL-12 therapies, which have shown potent antitumor efficacy but significant toxicity when administered systemically. The attIL12-T cells target tumor cells expressing cell-surface vimentin (CSV), which is often found on the surface of multiple tumor types. When these engineered T cells encounter tumor cells, they deliver IL-12 directly to the tumor microenvironment while minimizing systemic exposure. This localized delivery enriches effector T cells and interferon-gamma production in tumors, stimulates dendritic cell maturation, and activates secondary T-cell responses and tumor antigen spreading. Preclinical studies have demonstrated that attIL12-T cell therapy can eliminate peripheral cytokine release and associated toxic effects while maintaining remarkable antitumor efficacy in human and mouse large solid tumor models.
