attIL12-TIL is an autologous tumor-infiltrating lymphocyte (TIL) cell therapy engineered to express a membrane-anchored, tumor-targeted form of interleukin-12 (IL-12). Developed by the M.D. Anderson Cancer Center, this therapy specifically targets cell-surface vimentin (CSV), a marker frequently expressed on mesenchymal-derived tumor cells and associated with aggressive phenotypes in sarcomas. By anchoring IL-12 to the T cell membrane, the therapy aims to deliver the potent pro-inflammatory effects of the cytokine directly to the tumor microenvironment while minimizing systemic toxicity. This modification is designed to enhance T cell infiltration, overcome the collagen-rich physical barriers of high-grade tumors, and stimulate a robust anti-tumor immune response. It is currently being evaluated in Phase 1 clinical trials for advanced or metastatic soft tissue and bone sarcomas, including liposarcoma.
