Overview

ATTO-1310 is a novel, half-life extended, biparatopic anti-IL-31 antibody biotherapeutic developed using Attovia’s proprietary ATTOBODY platform. It is designed for subcutaneous administration and targets interleukin 31 (IL-31), a cytokine implicated in pruritic (itch-related) conditions. Each molecule consists of two VHH domains that bind distinct epitopes on IL-31, fused to a human IgG1-Fc engineered for extended half-life. This structure enables potent suppression of IL-31-mediated receptor dimerization and downstream signaling, resulting in rapid and sustained itch relief. The drug is being evaluated as a systemic treatment for chronic pruritus of unknown origin (CPUO) and atopic dermatitis, with the goal of infrequent dosing due to its long half-life[3][5][7][8]. Preclinical studies have demonstrated high affinity binding to IL-31, effective blockade of pruritus in animal models, favorable pharmacokinetics with >80% bioavailability after subcutaneous injection, low immunogenicity risk, and robust manufacturability from CHO cells[8].
