ATTR-01 is a genetically modified, oncolytic adenovirus type 5 (Ad5) engineered to specifically target and replicate within tumor cells that express the αvβ6 integrin. Upon selective infection of these tumor cells, ATTR-01 expresses a licensed anti-Programmed Death-Ligand 1 (PD-L1) human antibody, thereby delivering cancer immunotherapy directly within the tumor microenvironment. This approach aims to stimulate the patient's own immune system to attack and destroy the cancer. ATTR-01 is in early-stage clinical development for intravenous administration in adult patients with select epithelial solid tumors, including pancreatic, lung, bladder, head and neck, endometrial, and cholangiocarcinoma, which frequently express αvβ6 integrin[1][3][5]. The drug is under evaluation in Phase 1/2 trials using a master protocol design[1][3][8][9].
