Atu027 is a novel investigational RNA interference (RNAi) therapeutic designed to silence the expression of protein kinase N3 (PKN3) in the vascular endothelium. It consists of a chemically stabilized short-interfering RNA (siRNA) encapsulated in cationic and fusogenic lipids, forming a lipoplexed formulation. PKN3 acts as a downstream effector in the PI3K signaling pathway and is implicated in cellular morphology, locomotion, tumor progression, and metastasis formation. By targeting PKN3 mRNA via RNA interference, Atu027 inhibits local tumor invasion as well as lymph node and pulmonary metastasis in preclinical cancer models. The drug was developed primarily for advanced solid tumors and has been studied both alone and in combination with gemcitabine for advanced pancreatic carcinoma[1][4][6][7].
