Atuliflapon (AZD5718) is an oral, potent, and selective inhibitor of 5-lipoxygenase-activating protein (FLAP), developed by AstraZeneca. FLAP is a critical scaffold protein that facilitates the synthesis of leukotrienes from arachidonic acid. By inhibiting FLAP, atuliflapon prevents the production of pro-inflammatory lipid mediators, including leukotriene B4 (LTB4) and cysteinyl leukotrienes, which play significant roles in the pathophysiology of respiratory and cardiovascular diseases. The drug is primarily being investigated for its potential to reduce inflammation in patients with asthma, coronary artery disease (CAD), and chronic kidney disease (CKD). Clinical studies have focused on its ability to improve lung function in asthma and reduce biomarkers of vascular inflammation in cardiovascular settings.
