ATUX-1215 is a small molecule activator of Protein Phosphatase 2A (PP2A) currently under investigation for the treatment of high-risk neuroblastoma (NBL). It functions by reactivating PP2A, which subsequently leads to the dephosphorylation of MYCN, BRD4, and RNA Polymerase II. This mechanism suppresses MYCN expression and stability and modulates histone acetylation (specifically H3K27ac and H3ac). Research indicates that ATUX-1215 treatment upregulates the fibrinogen alpha chain (FGA) gene, a marker associated with improved overall survival in MYCN-amplified and stage 4 neuroblastoma patients.
