Overview

Atuzaginstat is an orally administered small molecule drug that acts as a first-in-class inhibitor of gingipain proteases, specifically targeting lysine gingipains (KGP) produced by the periodontal pathogen Porphyromonas gingivalis. It was developed to address neurodegenerative diseases, particularly Alzheimer's disease, based on evidence linking P. gingivalis infection and its proteases to Alzheimer’s pathology and inflammation. Atuzaginstat has also been investigated for periodontal disease and is in preclinical development for squamous cell cancer. The drug was originally developed by Cortexyme and later by Quince Therapeutics. In clinical trials, it showed some cognitive benefit in subgroups of Alzheimer’s patients with high P. gingivalis infection but failed to meet primary endpoints in the overall population; development has since been placed on clinical hold following these results[1][3][4][6][9].
