ATV-NDV is an **autologous tumor vaccine consisting of a patient’s own tumor cells modified with non-lytic Newcastle Disease Virus (NDV)**, sometimes further engineered to express immunomodulatory cytokines such as interleukin-7. This therapeutic vaccine is prepared by infecting autologous tumor cells with a lentogenic (low pathogenicity) NDV strain, followed by γ-irradiation to inactivate proliferative capacity, creating a live cell vaccine designed to stimulate the immune response against tumor-associated antigens. NDV imparts both **oncolytic (direct tumor-killing) activity and potent immunostimulatory effects**, serving as a biological adjuvant. The unique mechanism increases tumor immunogenicity and encourages robust anti-tumor T-cell activity—particularly by enhancing CD8+ cytotoxic T lymphocyte responses. ATV-NDV has been tested for adjuvant therapy post-surgical resection and shows promise for improving long-term survival in various malignancies, including melanoma, colorectal cancer, breast cancer, and glioblastoma[1][2].
