ATX-003

ATX-003 (also referred to as ATX3) is a localized pharmaceutical agent being developed for the treatment of chronic constipation, functional bowel disorders, and neurogenic bowel dysfunction, particularly for on-demand defecation in patients with spinal cord injuries. The drug was originally developed by Aayam Therapeutics using its proprietary LADR4 (Local Activation and Drug Release) platform, which is designed to create new chemical entities or novel formulations that provide targeted drug release to minimize systemic exposure. ATX-003 is formulated as a localized prodrug intended to trigger the rectocolonic reflex through cholinergic stimulation in the distal bowel. By acting specifically at the site of administration, it aims to provide a predictable, rapid bowel movement while avoiding the systemic side effects associated with traditional oral prokinetics. The compound is being developed through the FDA's 505(b)(2) regulatory pathway and was licensed to Dignify Therapeutics in 2023 for further clinical development.
