ATX-101 is a cell-penetrating peptide containing the APIM motif, designed to target proliferating cell nuclear antigen (PCNA) and disrupt protein complexes involved in DNA replication and repair. It has demonstrated antitumor activity in preclinical models of glioblastoma multiforme by reducing Akt/mTOR and DNA-PKcs signaling pathways[1][6]. Separately, another drug named ATX-101 refers to an injectable formulation of deoxycholic acid used for submental fat reduction; however, in the oncology context with this combination regimen, it refers to the APIM-peptide targeting PCNA[1][6]. Carboplatin is a platinum-based chemotherapy agent that causes DNA crosslinking and apoptosis. Pegylated liposomal doxorubicin (marketed as Caelyx) is an anthracycline antibiotic encapsulated in PEG-coated liposomes for improved delivery and reduced toxicity; it intercalates into DNA and inhibits topoisomerase II[7]. This combination is under investigation primarily for cancer treatment.
