Overview

ATX-295 is a novel, orally administered, selective small molecule inhibitor of KIF18A, a mitotic kinesin motor protein essential for cell division in certain tumors with chromosomal instability. By inhibiting KIF18A, ATX-295 disrupts chromosomal alignment and spindle microtubule dynamics during mitosis, leading to mitotic arrest and cell death specifically in cancer cells exhibiting aneuploidy or whole genome doubling. Preclinical studies have demonstrated robust anti-tumor activity in models of high-grade serous ovarian cancer and triple negative breast cancer. The drug is being developed by Accent Therapeutics and has received FDA Fast Track designation for the treatment of adult patients with advanced/metastatic platinum-resistant or refractory ovarian cancer. It is currently being evaluated in a first-in-human Phase 1/2 clinical trial enrolling patients with advanced solid tumors including ovarian cancer[1][2][3][4][6].
