Overview

ATYR2810 is a fully humanized monoclonal antibody developed by aTyr Pharma. It is designed to specifically and functionally block the interaction between neuropilin-2 (NRP2) and one of its primary ligands, vascular endothelial growth factor (VEGF), by directly binding at the VEGF binding pocket on NRP2. NRP2 is a cell surface receptor highly expressed in certain tumors, lymphatic tissue, and key immune cells implicated in cancer progression. Increased expression of NRP2 correlates with poor outcomes in many cancers due to its role in tumor invasiveness, metastasis, and therapy resistance. Preclinical studies have shown that ATYR2810 demonstrates anti-tumor activity as both monotherapy and in combination with chemotherapy or checkpoint inhibitors across several aggressive cancer models—including glioblastoma multiforme (GBM), triple-negative breast cancer (TNBC), non-small cell lung cancer, and clear cell renal cell carcinoma—by inhibiting tumor growth, reducing metastasis, downregulating genes linked to drug resistance (such as CXCR4), modulating immunosuppressive myeloid cells within the tumor microenvironment, and enhancing sensitivity to other therapies[3][4][5][6][7][8].
