Overview

Au-TMP is an experimental nanoparticle-based radiosensitizer consisting of gold (Au) nanoparticles conjugated with a tumor-homing peptide (TMP), specifically the TKPRP sequence. Developed by West China Hospital of Sichuan University, the gold nanoparticle core functions as a high-atomic-number (high-Z) material that enhances the efficacy of radiotherapy by increasing the local absorption of ionizing radiation and promoting the generation of reactive oxygen species (ROS). This process leads to targeted DNA damage and apoptosis within tumor cells. The TMP peptide (TKPRP) specifically targets Neuropilin-1 (NRP-1), a receptor frequently overexpressed in melanoma and tumor-associated vasculature, facilitating selective accumulation and cellular internalization of the nanoparticles. Au-TMP is administered via intratumoral injection and is currently being evaluated in Phase I clinical trials for the treatment of advanced melanoma in combination with radiotherapy and the anti-PD-1 antibody toripalimab.
