Auceliciclib (AU3-14) is a brain-penetrant, oral, small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6)[5][1][3][10]. It is developed by Aucentra Therapeutics and the University of South Australia as a targeted antineoplastic therapy primarily for glioblastoma (GBM), an aggressive brain malignancy with limited therapeutic options[5][1][9][3]. AU3-14 works by selectively inhibiting CDK4/6, key regulators of the cell cycle, leading to cell cycle arrest at G1/G0, induction of senescence, and anti-tumor activity[1][5]. In preclinical models and early clinical development, it efficiently crosses the blood-brain barrier, demonstrates substantial anti-tumor efficacy and safety, and shows a low risk of hematological toxicities such as neutropenia compared to other CDK4/6 inhibitors[1][7]. AU3-14 has also shown potential in combination with temozolomide, enhancing its efficacy and delaying resistance[1]. Clinical trials are ongoing or planned for glioblastoma, colorectal, and ovarian cancers[3][9].
