AUG-803

AUG-803 is an oral small molecule formulation of L-fucose being developed by AUG Therapeutics for the treatment of Leukocyte Adhesion Deficiency Type II (LAD II), a rare congenital disorder of glycosylation (CDG). LAD II is characterized by a defect in the transport of fucose into the Golgi apparatus, which results in the failure to synthesize fucosylated glycans such as sialyl-Lewis X. This impairment leads to severe immunodeficiency due to the inability of leukocytes to roll and adhere to the vascular endothelium. AUG-803 acts as a monosaccharide substrate replacement therapy, replenishing metabolic intermediates to bypass the transport defect and restore the synthesis of critical fucosylated glycoproteins. The drug was acquired from Avalo Therapeutics in September 2023 and is currently in early-stage clinical development.
