A combination therapy consisting of aumolertinib, a third-generation EGFR tyrosine kinase inhibitor, and pemetrexed, an antimetabolite chemotherapy agent. This combination has demonstrated sequence-dependent synergistic effects in EGFR-mutant non-small cell lung cancer (NSCLC), with administration of pemetrexed followed by aumolertinib showing the strongest anti-tumor and anti-metastasis effects. The synergy appears to involve enhanced suppression of the EGFR pathway and reduction in VEGF secretion, leading to normalization of intratumor blood vessels and increased intratumoral accumulation of pemetrexed. Clinical trials, including a phase II trial and the AENEAS2 trial, have shown promising efficacy results such as high objective response rates and prolonged progression-free survival compared to monotherapy, although with a higher incidence of adverse events like decreased neutrophil count.
