**AuNP-BBN-Pt1** is an investigational, non-PEGylated gold-nanoparticle theranostic designed for targeted treatment of gastrin-releasing peptide receptor-positive tumors, particularly prostate cancer. The nanoconstruct comprises a small gold nanoparticle functionalized with a thiolated DOTA derivative, the GRPR-targeting bombesin analog BBN[7-14], and a platinum(IV) prodrug. Bombesin-mediated GRPR binding promotes selective uptake by GRPR-positive cancer cells, while intracellular activation of the platinum prodrug is intended to produce platinum-mediated DNA damage and cytotoxicity. In PC3 prostate-cancer cells, AuNP-BBN-Pt1 showed cytotoxicity comparable with cisplatin and greater selectivity over non-tumoral RWPE-1 prostate cells; a separately radiolabeled 67Ga-AuNP-BBN-Pt1 derivative supported SPECT imaging and tumor-retention studies in a PC3 xenograft model.
