AUP-55 is an oncolytic bacteria-based Advanced Therapy Medicinal Product (ATMP) developed by Aurealis Therapeutics for oncology indications, primarily ovarian cancer and peritoneal carcinomatosis. It consists of genetically engineered *Lactococcus cremoris*, a non-pathogenic lactic acid bacterium, designed to deliver multiple therapeutic proteins directly into tumors via intratumoral injection. The mechanism of action is multi-faceted: - Activation of the innate immune system through bacterial membrane components acting as Toll-like receptor 2 (TLR2) and Nucleotide-binding oligomerization domain-containing protein 2 (NOD2) agonists. - Activation of the adaptive immune system via cytokines such as interleukin 15 (IL-15), interleukin 12 (IL-12), interleukin 18 (IL-18), granulocyte-macrophage colony stimulating factor (GM-CSF), and interferon alpha (IFN-A). - Blockade of angiogenesis and direct tumor cell killing through interferon alpha activity. The product acts as a live biopharmaceutical/gene therapy platform that induces tumor regression and improves survival in preclinical models for ovarian and intraperitoneal cancers[1][2][5].
