AUR108 is an investigational small molecule inhibitor of Interleukin-1 receptor-associated kinase 4 (IRAK4), currently being developed by Aurigene Discovery Technologies. IRAK4 is a critical component of the MyD88 signaling pathway; mutations in the MYD88 gene (such as L265P) lead to constitutive activation of IRAK4, which in turn drives the survival and proliferation of malignant B-cells through the NF-κB and MAPK pathways. These mutations are highly prevalent in certain B-cell malignancies, including activated B-cell diffuse large B-cell lymphoma (ABC DLBCL) and Waldenström's macroglobulinemia. AUR108 is designed to selectively inhibit IRAK4 activity to disrupt this oncogenic signaling cascade. It is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and preliminary efficacy in patients with relapsed or refractory advanced lymphomas, particularly those with MYD88 mutations.
