Overview

AURKA shRNA lentiviral transduction is an experimental gene therapy approach used to downregulate the expression of Aurora Kinase A (AURKA). In the context of glioblastoma research, this method involves using a lentiviral vector to deliver short hairpin RNA (shRNA) that specifically targets and degrades AURKA mRNA via the RNA interference (RNAi) pathway. AURKA is a key component of the ciliary disassembly complex; its inhibition or knockdown leads to the stabilization and increased abundance of primary cilia. Research suggests that restoring primary cilia in glioblastoma cells can significantly alter the cellular transcriptome, potentially suppressing multiple pro-cancer and immunosuppressive pathways. This approach is currently being explored as a ciliotherapeutic strategy to address the genetic complexity and immune evasion characteristic of malignant primary brain tumors.
