Overview
Aurothioglucose is a gold-containing compound formerly used as a disease-modifying antirheumatic drug (DMARD) for the adjunctive treatment of early active adult and juvenile rheumatoid arthritis not adequately controlled by other anti-inflammatory agents. It was also used in Felty's syndrome and psoriatic arthritis. The mechanism of action is not fully elucidated but is thought to involve inhibition of adenylyl cyclase activity in human lymphocyte membranes, which may reduce pro-inflammatory processes such as mast cell degranulation, histamine release, respiratory burst effects, macrophage activation, phagocyte induction/activation, and neutrophil chemotaxis. Aurothioglucose was administered via intramuscular injection; absorption from the injection site is slow and erratic. The drug has been discontinued in many markets due to safety concerns and the availability of more effective therapies[1][2][5][6].
