Drug intelligence / Profile preview
AUT1 (also known as RE01) is a first-generation, small-molecule positive allosteric modulator (PAM) of the voltage-gated potassium channel subtypes Kv3.1 and Kv3.2 (specifically Kv3.1b and Kv3.2a). Developed by Autifony Therapeutics, AUT1 acts by shifting the voltage-dependence of channel activation toward more hyperpolarized potentials, thereby facilitating channel opening and enhancing the high-frequency firing fidelity of fast-spiking parvalbumin-positive (PV+) GABAergic interneurons. It has been widely utilized as a preclinical research tool to investigate the therapeutic potential of Kv3 channel modulation in central nervous system disorders. Preclinical studies have demonstrated its efficacy in animal models of schizophrenia (reversing cognitive deficits), bipolar disorder/mania (reducing amphetamine-induced hyperactivity), and Alzheimer's disease (rescuing deficits in interneuron firing and gamma-oscillations).
