AUTO-5 is an autologous CAR-T cell therapy developed for the treatment of peripheral T-cell lymphoma. It specifically targets TRBC2 (T cell receptor beta constant 2) on malignant T cells using a chimeric antigen receptor (CAR) approach. The therapy is designed to selectively eliminate cancerous T cells while preserving a portion of the patient's normal, healthy T cells to maintain immune function. A safety switch (RQR8) is incorporated into the product to allow physicians to manage toxicity by eliminating the programmed T cells if severe adverse effects occur. The drug was initially developed by University College London and is being advanced by Autolus Ltd. As of March 2025, it remains in preclinical development.[1][2][3]
