Overview

AUTO2 is a dual-targeting CAR-T cell therapy developed by Autolus for the treatment of multiple myeloma. It is designed to recognize and attack cancer cells expressing both B-cell maturation antigen (BCMA) and transmembrane activator and CAML interactor (TACI), using a natural human ligand (APRIL) that binds to both receptors. This approach aims to address the challenge of variable BCMA expression on myeloma cells, potentially reducing relapse due to loss of BCMA expression. The therapy also incorporates an RQR8 safety switch, which allows for rapid elimination of the CAR-T cells with rituximab in case of severe side effects. AUTO2 was administered following pre-conditioning chemotherapy, and its development included clinical trials focused on determining safety, dosing, and efficacy in multiple myeloma patients. However, development was discontinued after initial studies due to lack of differentiation from competitor therapies.
