AUTO4 is an autologous chimeric antigen receptor (CAR) T-cell therapy in clinical development for the treatment of T-cell lymphomas, specifically targeting cancers derived from the T cell receptor constant region beta chain 1 (TRBC1). Approximately 40% of T cell lymphomas express TRBC1, making this a significant target population. AUTO4 is engineered to coexpress a humanized second-generation CAR that targets TRBC1 and includes the RQR8 safety switch, which allows selective depletion of transgenic T cells if needed. The therapy is being developed by Autolus Therapeutics and has shown promising results in early-phase clinical trials for relapsed or refractory peripheral T-cell lymphoma (PTCL), with evidence of durable responses and a favorable safety profile.
