AUTO6 is a second-generation, GD2-targeting chimeric antigen receptor T cell (**CAR T**) therapy developed as a potential treatment for neuroblastoma and other GD2-positive solid tumors. The construct was designed to avoid on-target, off-tumor neurotoxicity associated with GD2, a surface antigen broadly expressed on neuroblastoma cells, by using an optimized GD2 binder. AUTO6 induces clinical activity, causing rapid regression of solid tumor burden, and has demonstrated safety by avoiding neurotoxicity that can be seen with anti-GD2 antibody therapies. AUTO6 was developed through a collaboration between Autolus Therapeutics and University College London, with early phase trials sponsored by Cancer Research UK’s Centre for Drug Development. Its ongoing development targets relapsed or refractory childhood neuroblastoma, an area of substantial unmet need[1][3].
