AUTO6NG is a next-generation programmed T cell therapy (CAR-T cell therapy) designed to target GD2-expressing cancer cells. It builds upon the earlier AUTO6 product by incorporating additional cell programming modules that help overcome immune suppressive defense mechanisms in the tumor microenvironment and enhance CAR T cell persistence. The primary mechanism involves genetically engineered T lymphocytes expressing a chimeric antigen receptor (CAR) specific for Disialoganglioside GD2, an antigen commonly found on neuroblastoma and other solid tumors. AUTO6NG is being developed primarily for the treatment of relapsed or refractory neuroblastoma in pediatric patients but is also under investigation for other GD2-positive solid tumors such as osteosarcoma, melanoma, soft tissue sarcoma, small cell lung cancer (SCLC), and H3K27M mutant diffuse midline glioma. The drug is currently in Phase 1 clinical trials.
