Drug intelligence / Profile preview
BNT122 + atezolizumab + fluorouracil + leucovorin + irinotecan + oxaliplatin is an investigational combination immunotherapy and chemotherapy regimen being evaluated for the adjuvant treatment of resected pancreatic ductal adenocarcinoma (PDAC). The regimen is built around autogene cevumeran (BNT122; RO7198457), an individualized, mRNA-based neoantigen cancer vaccine (iNeST) co-developed by BioNTech and Genentech. Autogene cevumeran is a uridine mRNA-lipoplex designed to encode up to 20 patient-specific tumor neoantigens to stimulate a targeted T-cell response. This vaccine is combined with atezolizumab (Tecentriq), an anti-PD-L1 monoclonal antibody checkpoint inhibitor, and mFOLFIRINOX, a standard cytotoxic chemotherapy regimen consisting of 5-fluorouracil, leucovorin, irinotecan, and oxaliplatin. The combination is currently being evaluated in the randomized Phase II IMCODE003 trial (NCT05968326) sponsored by Genentech, comparing the regimen to mFOLFIRINOX alone.
