Autogene cevumeran + pembrolizumab is a combination therapy under clinical investigation for the treatment of advanced melanoma and possibly other solid tumors. **Autogene cevumeran** is a personalized cancer vaccine based on uridine mRNA lipoplex technology; it is designed using individual patient's tumor-specific somatic mutation data to encode up to 20 patient-specific neoantigens, stimulating both CD4^+^ and CD8^+^ T-cell immune responses against the tumor. **Pembrolizumab** is a monoclonal antibody immune checkpoint inhibitor that targets the programmed cell death protein 1 (PD-1) receptor, thereby augmenting anti-tumor immunity by blocking inhibitory signals to T cells. The combination aims to enhance antitumor efficacy by combining mRNA neoantigen vaccination with PD-1 blockade, promoting both the generation and function of tumor-specific T cells. The two agents are administered together in a clinical setting to evaluate efficacy, safety, pharmacokinetics, and patient-reported outcomes, particularly in previously untreated advanced melanoma patients[2][3][4][5].
