Overview

Autologous AdE1-latent membrane protein CTLs are a form of adoptive cell therapy in which a patient's own cytotoxic T lymphocytes (CTLs) are expanded ex vivo to specifically target latent membrane proteins (primarily LMP1 and/or LMP2) encoded by the Epstein-Barr virus (EBV). These proteins are expressed in certain EBV-associated malignancies, such as nasopharyngeal carcinoma and some lymphomas. The process typically involves stimulating patient-derived peripheral blood mononuclear cells with antigen-presenting cells expressing non-toxic or mutant forms of the viral latent membrane proteins—sometimes using adenoviral vectors like AdE1-LMPpoly—to generate polyclonal populations of functional, tumor-targeting T cells. These expanded CTLs can then be infused back into the patient to mediate anti-tumor effects by recognizing and killing EBV-infected malignant cells that express these antigens. Clinical studies have shown promising results for this approach in patients with relapsed or refractory EBV-associated cancers[2][5][6].
