The autologous AdHER2 dendritic cell vaccine is an investigational personalized cancer immunotherapy designed to target tumors expressing the human epidermal growth factor receptor 2 (HER2). The therapy involves isolating a patient’s own mononuclear cells via apheresis and generating dendritic cells (DCs) that are then transduced ex vivo with an adenoviral vector encoding the extracellular and transmembrane domains of human HER2 (AdHER2). The product is manufactured under Good Manufacturing Practice regulations at specialized centers such as the NIH Clinical Center[1][6]. Primary indications under investigation include metastatic solid tumors with HER2 expression—including breast, ovarian, stomach/colon, bladder, and endometrial cancers—especially in patients who have progressed after standard therapies[1][4][6].
