Overview

Autologous anti-CD123 CAR-T cells are genetically engineered T-cell therapies designed to target CD123 (interleukin-3 receptor subunit alpha), which is highly expressed on various hematological malignancies, including acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), and blastic plasmacytoid dendritic cell neoplasm (BPDCN). The therapy involves harvesting a patient's own T cells, modifying them ex vivo with a chimeric antigen receptor (CAR) construct targeting CD123 (typically utilizing a costimulatory domain such as 4-1BB or CD28 and a CD3ζ signaling domain), and reinfusing them into the patient. Beijing Boren Hospital and other institutions have evaluated autologous anti-CD123 CAR-T cells in clinical trials for relapsed or refractory CD123-positive hematological malignancies.
