Autologous anti-EpCAM CAR-T cells is an investigational autologous chimeric antigen receptor T-cell (CAR-T) therapy targeting epithelial cell adhesion molecule (EpCAM). It is being developed by Shanghai 10th People's Hospital, Biomed Union Biotechnology, and Shanghai Ultra-T Immune Therapeutics for the treatment of EpCAM-positive recurrent or refractory advanced solid tumors, including gastrointestinal cancers (such as pancreatic, colorectal, and gastric cancers), lung cancer, and mesothelioma. The CAR-T cells are genetically engineered to express a CAR specific for EpCAM, allowing them to selectively bind and eliminate EpCAM-expressing tumor cells. The construct also incorporates a safety switch based on truncated epidermal growth factor receptor (tEGFR) to mitigate potential toxicities like cytokine release syndrome (CRS).
